So I need to reinstall the dev build for TMP to work with FF3.0.1, but I don't know if this will delete my current preferences, and if so, how can I keep them or back them up?

You must not worry about that, all preferences are kept with re-install or uninstall.

If you want export/import preferences, click [Tools] - [Tab Mix Plus Options] [Settings] button, choose 'Import Settings' or 'Export Settings'.
